Synthesis of Novel Building Blocks for Peptidomimetics

Peptidomimetics are unnatural oligomers which mimic protein secondary structures by folding into well-defined three-dimensional structures. They display a protein-like functionality without labile amide linkage.

An interesting approach is to replace the amide linkage with 1,2,3-triazole rings in order to build triazolamers (Fig 1). Some triazolamers which adopt zigzag conformations reminescent of peptide β-strands have been reported. Computational and experimental studies suggest that triazolamers offer attractive starting points for the design of protease inhibitors.

The aim of this project is to synthesize several triazolamers with a wide variety of side chain functional groups in order to explore the scope of these extremely interesting compounds.
